Purpose:
Myricetin, a common dietary flavonoid, is widely distributed in fruits and vegetables and is used as a health food supplement based on immune function, antioxidant, antitumor and anti-inflammatory properties. The aim was to investigate the effect of myricetin in combination with radiation therapy to increase radiation sensitivity in lung cancer A549 and H1299 cells.

Methods:

A549 cells and H1299 cells were exposed to X-ray with or without miricetin treatment. Colony formation assays, in the CCK-8 assay, by flow cytometry and caspase-3 leveling were used to evaluate the miricetin radiosensitizing activity for cell proliferation and apoptosis in vitro. The nude mouse tumor xenograft model was built to evaluate the effect of radiation sensitization with Myricetin in vivo.

Results:
Comparing the exposed group without miricetin treatment, the treated group had significantly miricetin suppressed survival fraction and cell proliferation, increased cell apoptosis and increased caspase-3 protein expression after X-ray exposure in vitro. And in an in vivo study, the tumor growth rate of xenografts was significantly reduced in mice treated with miricetin.

Conclusions:
The study showed both in vitro and in vivo evidence that the combination of Myricetin radiotherapy may increase tumor radiosensitivity in pulmonary H1299 carcinoma and A549 cells, and myricetin may be a potential radiosensitizer in lung cancer therapy.

The stimulatory neurotransmitter glutamate can accumulate in the brain and is thought to be involved in the etiology of many neurodegenerative disorders, including ischemia and Alzheimer’s disease. Therefore, it is important to look for compounds that reduce glutamate neurotoxicity. This glutamate-mediated excitotoxicity is caused by intracellular Ca2 + overload through the N-methyl-D-aspartate receptor (NMDAR), reactive oxygen species (ROS) generation, and caspase-3 activation. Here, we show that the natural flavonoid myricetin inhibited glutamate-induced excitotoxicity and protected neurons by several different mechanisms. First, Myricetin affected modulation of NMDAR phosphorylation, leading to a subsequent reduction in glutamate-induced intracellular Ca2 + overload. Second, myricetin inhibited ROS-induced glutamate.Finally, glutamate-induced activation of caspase-3 decreased with miricetin treatment.
